摘  要:建立麦胚油中二十八烷醇的毛细管气相色谱含量分析方法。以氯仿作为溶剂,外标法定量。采用了DB-5ht毛细管柱(15m×250μm×0.10μm)和气相色谱仪(配FID检测器)。进样口温度350℃,检测器温度385℃,柱箱升温程序:初始温度120℃,维持1min后,以20℃/min升温至300℃,维持3min,再以10℃/min升温至380℃,维持10min,高纯氮气(载气)恒压15psi,氢气流速:40mL/min,空气流速:400mL/min,进样量为1μL。结果表明二十八烷醇浓度在0 300mg/L范围内呈良好的线性关系(相关系数达0.999以上);测试三个浓度的平均回收率在96.2%99.7%之间,RSD均少于5%;精密度(n=5)RSD为2.8%;方法检出限为1.3μg/mL。经证实,采用DB-5h毛细管柱(15m)能使麦胚油中的杂质和二十八烷醇得到良好分离。本法准确、灵敏、快速、重现性好,可用于麦胚油及其胶囊中的二十八烷醇定量分析。A gas chromatography method for the determination of octacosanal in wheat germ oil was developed.The solvent was chloroform and external standard method was applied.DB-5ht capillary column (15m ×250μm ×0.10μm)and gas chromatograph (with flame ionization detector) were used in the analysis.Injector temperature was 350℃,detector temperature was 385℃,and the column temperature was raised by automatic programming:initial temperature was 120℃,maintaining 1min,the temperature rising to 300℃ (20℃/min) for 3min,and then rising to the final temperature380℃℃ (10℃/min) for 10min.The constant pressure of nitrogen (carrier gas) with high purity was 15psi,the flow rate of hydrogen was 40 mL/min,and air was 400 mL/min.Injection volume was 1μL.The results showed in the ranges of 0 to 300mg/L of octacosanol,it had good linearity [the correlation coefficient (r) ≥0.999] ;Mean recoveries with three concentration tests were from 96.2 to 99.7%,and RSDs were all under 5% ; RSD was 2.8% ; MDL was 1.31μg/mL.It was proved that complete separation of impurity and octacosanal in wheat germ oil was obtained by a DB-5ht capillary column (with 15m).The method was suitable for quantitative analysis of octacosanal in wheat germ oil and wheat germ oil capsules.It is accuracy,sensitive,rapid and reproducible.
