银条多糖SFPSA的单糖组成及抗肿瘤活性研究

摘　　要：本文对银条多糖(SFPSA)的单糖组成和抗肿瘤活性进行了研究。采用高效液相凝胶渗透色谱法(HPGPC)测定其纯度和分子量分布。采用糖腈乙酸酯衍生化法分析SFPSA的单糖组成。运用MTT法考察SFPSA对三种肿瘤细胞株(BGC-823、HGC-27和HT-29)增殖的抑制作用。结果表明SFPSA是均一的多糖组分,其相对平均分子量为168.30 kDa;SFPSA中主要含鼠李糖、阿拉伯糖、葡萄糖和半乳糖等单糖组分,其摩尔百分含量比为8.19∶37.74∶3.46∶50.61;SFPSA对三种肿瘤细胞的增殖均有抑制作用,其中对人结肠癌HT-29细胞增殖抑制作用最为明显,其作用72 h后的半数抑制率为407.52μg/mL。因此,SFPSA具有良好的体外抗结肠癌活性。Monosaccharide composition and antitumor activities of acidic polysaccharide （SFPSA） from Stachysfloridana Schuttl.ex Benth were studied.The monosaccharide compositions of SFPSA were determined using sugar nitriles acylation method.The proliferation inhibitory activities of SFPSA against three tumor cell lines （BGC-823,HGC-27 and HT-29） were evaluated by MTT assay.The results showed that SFPSA with average molecular weight of 168.30 kDa was mainly composed of rhamnose,arabinose,glucose and galactose with the molar ratio of 8.19∶ 37.74∶ 3.46∶ 50.61.In addition,SFPSA can inhibit the proliferation of three cell lines in dose-and time-dependent manner.Human colon cancer HT-29 cell was more sensitive to SFPSA than BGC-823 and HGC-27,and the half inhibition concentration of SFPSA on HT-29 was 407.52 μg/mL after drug administration for 72 h.Therefore,it was concluded that SFPSA had a good in vitro anti-colon cancer activity.
