摘  要:目的:研究柯萨奇B4病毒(CVB4)jlu06株3A蛋白基因特征,探讨该株病毒与致病相关的分子基础。方法:RT-PCR扩增目的基因片段,测序后进行同源性、变异性及遗传进化分析。结果:CVB4jlu06株3A蛋白基因全长267个核苷酸,编码89个氨基酸;其基因序列与糖尿病分离株(Tuscany株)和标准株(JVB)同源性较高,该株病毒3A蛋白7个位点的氨基酸与非致糖尿病标准株不同。结论:CVB4jlu06株3A蛋白基因序列与Tuscany株和JVB株具有相同的进化途径,CVB4jlu06株3A蛋白不同于标准株的氨基酸变异可能是其致病表型的分子基础。Objective: To research the gene characteristics of Coxsackievirus B4 (jlu06 strain) 3 A protein, explore the pathogenic molecular basis of Coxsackievirus B4. Methods : The target gene fragment was amplified by RT - PCR, the homology analysis, mutation a- nalysis, and genetic evolution analysis were conducted after gene sequencing. Results: The total length of Coxsackievirus B4 (jlu06 strain) 3A protein included 267 nucleotides, which encoded 89 amino acids; its gene sequence showed high homology with diabetogenic strain (Tus- cany) and type strain (JVB), the nucleotide of seven loci in Coxsackievirus B4 (jlu06 strain) 3A protein were different from nondiabetogenic JVB strain. Conclusion: The evolution pathway of gene sequence of Coxsackievirus 134 (jlu06 strain) 3A protein is the same as Tuscany strain and JVB strain, the mutation of amino acids of Coxsackievirus B4 (jlu06 strain) 3A protein different from the type strain might be the molecular basis of its pathogenic phenotype.
