摘 要:[目的]检测初摘烟叶中直链淀粉和支链淀粉的含量。[方法]采用双波长分光光度法,选择直链淀粉检测波长为462和610 nm,支链淀粉检测波长为516和728 nm。根据标准样品的回归方程,可得到直链淀粉和支链淀粉的含量。[结果]直链淀粉的最大吸收波长为610 nm,支链淀粉的最大吸收波长为516 nm,在烟叶中含量分别为6.34%和20.41%。[结论]双波长分光光度法检测直链淀粉和支链淀粉操作简单,结果迅速,可用作批量检测。[ Objective ] To determine the contents of amylose and amylopectin in tobacco leaves. [ Method ] Dual wavelength spectrophotometry was used, and the measurement wavelengths of 462 and 610 nm for amylose, while 516 and 728 nm for amylopectin were selected accord- ing to the principle of dual wavelength calorimetric. The contents of amylose and amylopectin in tobacco" leaves were calculated based on re- gression 'equation. [ Result] The maximum absorption wavelength were 610 and 516 nm respectively for the two complexes, and the contents were 6.34% and 20.41%. [ Conclusion] The contents of amylose and amylopectin in tobacco leaves determined with dual-wavelength spec- trophotometry were accurate, and the method was suitable for the determination of amylose and amylopectin in tobacco leaves samples.
