摘  要:目的:研究柯萨奇B3病毒(CVB3)/MKP株5'非编码区基因序列及变异性,探讨其致病性的分子基础。方法:用Hela细胞培养柯萨奇B3病毒,RT-PCR法扩增5'非编码区,测序后进行同源性、变异性及遗传进化分析。结果:CVB3/MKP株5'非编码区基因全长742个核苷酸,该株病毒与CVB3/20、CVB3/28、CVB3/0、CVB3/Nancy、CVB3/AS、CVB3/Woodruff(CVB3/W)及CVB3(CXA3CG)株5'非编码区同源性在91.9%~99.6%之间,而与CVB3/GA和CVB3/CO两种病毒株同源性分别为83.6%和83.3%。CVB3/MKP株的5'非编码区序列与既致胰腺炎又致心肌炎的病毒株共同聚簇,而与非致病毒株和仅致胰腺炎的病毒株距离较远。结论:CVB3/MKP株5'非编码区基因序列可能与病毒的毒力表型有关。Objective: To study the gene sequence and vari- ation of 5 "untranslated region of Coxsackievirus B3 ( CVB3 ) /MKPstrain, explore the pathogenic molecular foundation. Methods: CVB3 was cultured in HeLa cells, RT - PCR was used to amplify 5"untranslated region of CVB3, and then homology analysis, variation analysis, and genetic evolution analysis were carried out after gene sequencing. Results: There were 742 nucleotides in 5"untranslated region of CVB3/MKP strain, the homologies of CVB3/MKP strain and CVB3/20, CVB3/28, CVB3/0, CVB3/Nancy, CVB3/AS, CVB3/ Woodruff (CVB3/W), CVB3 (CXA3CG) strains were 91.9% -99.6%, but its homologies with CVB3/GA and CVB3/CO were 83.6v/r and 83.3% , respectively. The 5"untranslated region of CVB3/MKP strain clustered together with virus strains indncing panereatitis and luyoearditis, hut it was apart from non - virulence strains and strains inducing pancreati- tis. Conclusion: The gene sequence of 5"untranslated region of CVB3/MKP strain is correlated with viral virulence phenotype.
