摘 要:目的从变黑雷松藻(Lessonia nigrescence)的根部和茎部中分别提取分离得到2种褐藻胶(LNA1,LNA2)和2种褐藻糖胶(LNF1,LNF2),在对其进行理化性质和结构分析基础上,进一步对其抗凝活性进行评价。方法运用醋酸纤维素膜电泳(CME)、高效凝胶渗透色谱(HPGPC)、高效液相色谱(HPLC)对4种多糖进行纯度、分子量和单糖组成分析;采用红外光谱(IR)和核磁共振氢谱(1 H-NMR)对4种多糖进行结构分析;通过活化部分凝血活酶时间(APTT)和凝血酶原时间(PT)评价4种多糖抗凝活性。结果根部褐藻胶中甘露糖醛酸含量(73%)明显高于颈部(61%),LNF1和LNF2均是以岩藻糖、木糖和半乳糖为主的硫酸岩藻聚糖,均能显著延长APTT,但对PT影响较小。结论变黑雷松藻根部褐藻胶中甘露糖醛酸含量明显高于茎部,褐藻糖胶中的硫酸酯基主要存在于岩藻糖的C4和C2/C3位,且主要通过影响内源凝血途径发挥抗凝作用。objective To perform a comparative study of the structures and anticoagulant activities of the extracted alginates (LNA1, LNA2 ) and fucoidans (LNF1 and LNF2) from the roots and the stems of Lessonia nigrescence. Methods Physicochemical properties of four kinds of polysaccharides were studied by cellulose-acetate membrane electrophoresis (CME), high performance gel permeation chromatogra- phy (HPGPC) and high performance liquid chromatograph (HPLC), structural comparisons among them were separately analyzed by infrared spectroscopy (IR) and ^1H-NMR. The anticoagulant activites were evaluated by activated partial thromboplastin time (APTT) and prothrombin time (PT). Results The mannuronic acid contents of LNA1 (73 %) were significantly higher than that of LNA2 (61%), while the fucoidans I.NF1 and LNF2 were mainly composed by fucose, xylose and galactose, and the anticoagulant effect demonstrated that LNF1 and LNF2 could significantly prolong the APTT but less effect on PT. Conclusion The alginates from the roots (LNA1) were composed of higher mannuronic acid than that from the stems (LNA2). The sulfate substitution positions were located at C4 and C2/C3 of fucose residues of LNF1 and LNF2, and the anticoagulant effects were mainly through the endoge-nous coagulation pathway.
