摘  要:目的建立食品中L-抗坏血酸、D-异抗坏血酸及抗坏血酸总量的高效液相色谱法。方法试样经偏磷酸溶液超声提取后,液相色谱法直接测定还原型L-抗坏血酸和D-异抗坏血酸;加入L-半胱氨酸溶液,将脱氢抗坏血酸还原为还原型抗坏血酸后测定L-抗坏血酸总量。流动相为磷酸二氢钾-十六烷基三甲基溴化铵-甲醇(pH2.5~2.8);检测波长245 nm,外标法定量。结果 L-抗坏血酸和D-抗坏血酸在0.5μg/ml^50μg/ml范围内线性关系良好;平均添加回收率为84.4%~110.8%,RSD<7.00%(n=6);固体样品的定量限为3.125 mg/100 g,液体样品的定量限为0.625 mg/100 g。结论该方法简便、快速、准确,能够检测食品中L-抗坏血酸、D-异抗坏血酸及L-抗坏血酸总量。Objective To establish an HPLC method for the analysis of L-ascorbic acid,D-isoascorbic acid and total ascorbic acid in food.Methods After ultrasonic extraction of the sample in metaphosphoric acid solution,L ascorbic acid and D-ascorbic acid were analyzed at 245 nm by liquid chromatography using 98% potassium dihydrogen phosphate(0.05 mol / L)-cetyl trimethyl ammonium bromide-2% methanol as the mobile phase.Total L-ascorbic acid was analyzed after reduction of dehydro-L(+) ascorbic acid with cysteine under the same chromatographic condition.Results L-ascorbic acid had a good linerility in the range of 0.5 μg / ml ~ 50 μg / ml with recoveries of 84.4% ~ 110.8% and RSD 7.00%(n = 6),and the limits of quantitation(LOQ) were 3.125 mg /100 g and 0.625 mg /100 g in solid and liquid sample,respectively.Conclusion The method is simple,quick and accurate and suitable for the determination of L-ascorbic acid,D-isoascorbic acid and total L-ascorbic acid in foods.
