摘 要:目的:探讨茶多酚能否诱导鼻咽癌细胞系 CNE1-LMP1凋亡及凋亡发生的分子机制。方法:用不同浓度的茶多酚处理 CNE1-LMP1细胞,分别用荧光显微镜、 DNA琼脂糖凝胶电泳、 MTT和流式细胞仪等方法,观察细胞生化和形态学改变;同时,确定茶多酚诱导细胞凋亡过程中 caspase-3活性的动力学变化。结果:茶多酚处理后,荧光显微镜下可见 CNE1-LMP1细胞在形态学上出现细胞核固缩、核碎裂、凋亡小体等凋亡细胞的形态学改变;琼脂糖凝胶电泳 DNA呈典型的“梯状”条带; MTT测定发现茶多酚处理后细胞生长受到不同程度的抑制。同时, CNE1-LMP1细胞在凋亡过程中 caspase-3活性的升高,呈明显的时效关系。结论:茶多酚能诱导 CNE1-LMP1细胞凋亡,可能通过 caspase-3的活化诱导细胞凋亡。Objective: The current study was designed to investigate the apoptosis induced by tea polyphenols in nasopharyngeal carcinoma cell line CNE1-LMP1 and molecular mechanism in the apoptotic process. Methods: After treatment of CNE1-LMP1 cells with various concentrations of tea polyphenols,apoptosis of the cells was detected by fluorescent microscopy, DNA gel electrophoresis, MTT assay and flow cytometry, and the activity of caspase-3 was detected by Western blot analysis and caspase-3 assay at different time points. Results:Typical nuclear condensation was observed under fluorescent microscopy, DNA Ladder appeared on the 2% agarose gel. Survival rate of the cells was decreased. Furthermore, the activity of caspase-3 induced by tea polyphenols in time-dependent manner during apoptotic process. Conclusion: Tea polyphenols could induce apoptosis of CNE1-LMP1 and the activity of caspase-3 might be involved in the apoptotic process.
