摘 要:应用响应面分析法优化甘蔗糖厂滤泥中叶绿素提取工艺。在单因素试验基础上,选取提取温度、提取时间、料液比为自变量,叶绿素的提取量为响应值,根据Box-Behnken中心组合试验设计原理采用3因素3水平的响应面分析法,研究各自变量及其交互作用对叶绿素提取量的影响,模拟得到二次多项式回归方程的预测模型。结果表明,甘蔗糖厂滤泥提取叶绿素的最优提取工艺参数为:提取温度32℃、提取时间2 h、料液比1︰7(g/mL),在此条件下叶绿素提取量的实际值为0.192 mg·g-1,理论值为0.189 mg·g-1,两者的相对误差为1.59%,说明响应面法优化甘蔗糖厂滤泥中叶绿素提取工艺的可行性和合理性。The conditions of extracting chlorophyll from filter mud in sugar cane mill factories were optimized based on Response Surface Methodology. Three extraction parameters including extraction temperature, extraction time and ratio of material to liquid were optimized using central composite design and response surface methodology based on single factor investigations for achieving maximum the chlorophyll extraction quantity. The interaction of the respective variables and their influence on the chlorophyll exlraction yield were studied by using Box-Behnken cenWal composite design and response surface analysis theory, and the simulated quadratic polynomial regression equation of prediction model was set up. The results showed that the optimal condition for extraction of chlorophyll was that the extraction temperature of 32 ℃, the extraction time of 2 h and the material to liquid ratio of 1 : 7 (g/mL). Under this condition, the measured extraction yield of chlorophyll was 0.192 mg.g-1, and the relative error was 1.59% compared with the theoretical value of 0.189 mg.g-1. It is feasible and reasonable that optimization of the extraction technology for chlorophyll from filter mud in sugar cane mill factories by response surface methodology.
