摘 要:优化了同时检测岩藻糖等7种单糖和海藻糖标准品的高效阴离子交换色谱一脉冲安培检测条件;研究了提取方式、温度和时间对食用茵中单糖和海藻糖提取效率的影响;最后检验方法性能,确立了适于食用菌单糖分离和检测的方法。结果表明,高速均质2min提取食用茵鲜样或60℃超声60min提取食用菌干样时,单糖提取效率最高。离子色谱检测条件为:CarboPacMAl色谱柱,流动相为去离子水:600mMNaOH溶液=20:80(体积比)。8种糖的最低检出限为5.5—63.6μg/L,食用菌鲜样和干样中的定量限分别为0.37—4.24mg/Kg和3.68—42.39mg/Kg。方法加标回收率为76.1%~127.1%,相对标准偏差1.44%~8.43%。A high performance anion-exchange chromatography-pulsed amperometric detection (HPAEC-PAD) method was developed and optimized for the analysis of seven standard preparations of monosaecharides and trehalose. In addition, the effects of extraction parameters on extraction of monosaeeharides and trehalose in edible fungi were investigated with the developed HPAEC-PAD method. The results showed that, the extraction efficieneies were the highest when the extraction methods were homogenized with high speed for 2 min for fresh Lentinus edodes and fresh Agaricus bisporus, and ultrasonic extraction for 60 min at 60℃ for dry Ganoderma lucidum and dry Auricularia auricula respectively. The parameters of chromatographic detection were optimized as follows : the chromatographic column was CarboPae MA1, and the eluting phases were deionized water:600 mM NaOH solution = 20:80 ( V: V). The limits of detection were 5.5 μg/L-63.6 μg/L,and the limits of quantitation were 0.37 -4.24 mg/Kg for fresh mushroom and 3.68-42.39 mg/Kg for dry mushroom respectively. Moreover, the recovery rates of the method were 76. 1%-127. 1% and the RSDs were 1.44% -8.43% based on the parameters above.
