茅台酒与乙醇影响二乙基亚硝胺致小鼠肝脏MMP2表达的差异

摘　　要：目的:研究茅台酒与乙醇影响二乙基亚硝胺(DEN)致小鼠肝脏金属基质蛋白酶2(MMP2)表达的影响。方法:将54只C57BL/6J小鼠随机分为正常对照组、茅台酒干预组、乙醇干预组,分别给予无菌生理盐水、茅台酒和乙醇灌胃处理,茅台酒干预组和乙醇干预组用DEN腹腔注射,35周后取小鼠肝脏,采用实时荧光定量PCR(RT-PCR)、免疫组化染色及Western blot检测肝组织内MMP2 mRNA及蛋白的表达。结果:MMP2基因在乙醇干预组中的mRNA表达显著高于正常对照组、茅台酒干预组,差异有统计学意义(P<0.05);MMP2 mRNA水平在茅台酒干预组与正常对照组之间的差异无统计学意义(P>0.05);乙醇干预组MMP2蛋白表达比茅台酒干预组高,差异有统计学意义(P<0.05)。结论:乙醇协同DEN致小鼠肝脏MMP2表达上调,含同样乙醇浓度的茅台酒没有这种作用。Objective： To study the impact of Maotai liquor and ethanol on the expression of matrix metalloproteinases 2 （MMP2） in liver of mice treated by DEN. Methods： C57BL/6J mice were even- ly and randomly allocated into 3 groups with 18 mice in each （Maotai liquor treating group, ethanol treating and control group）. After 35 weeks, the liver tissues of all groups were collected, and the MMP2 expression in liver tissues was probed by real-time PCR, western blot and immunohistochemistry methods. Results： Our data showed that the MMP2 mRNA expression of ethanol treating group was higher than that of Maotai liquor treating group and control group with statistical significance （P 〈 0.05 ） ; However, the difference of the expression of MMP2 mRNA between Maotai treating and control group was not evident（P 〉0.05） ;The protein expression of MMP2 displayed similar pattern to that of mRNA. The MMP2 protein expression level in ethanol treating group was higher than that of Maotai treating group （P 〈 0.05 ）. Conclusions： Ethanol solution, but not Maotai liquor （ with equal content ethanol）, can collaborate DEN on the increase of MPP2 expression in mice liver tissue.
