摘 要:利用实验室保藏菌种白腐真菌发酵产木聚糖酶,采用麦麸、玉米秸秆及半纤维素作为诱导物,发现诱导物的诱导作用明显,其中以玉米秸秆的诱导效果最为显著,其最佳添加量为2 g/L,酶活达到11.8 U/mL,比不添加诱导物提高了61.64%。在单因素试验的基础上,采用正交设计法对发酵产酶培养基条件进行优化,实验结果证明,最佳营养基质和配比为酵母膏6g/L,硫酸铵2g/L,氯化钠3g/L,V B10.01 g/L,pH 6.0,在此基础上获得的最大酶活为21.15 U/mL,比优化前提高了79%。White rot fungi preserved in our laboratory was used to obtain xylanase. Wheat bran, corn stalk and hemicellulose were used to as inducer to improve xylanase activity. The results showed that corn stalk was the best inducer. When the corn stalk addition was 2 g/L, the xylanase activity reached to 11.8 U/mL and increased by 61.64 %. On the basis of single of factor experiments, orthogonal design method was used to optimize the enzyme producing medium. The results showed that the best addition of yeast extract, ammonium sulphate, sodium chloride and VB, was 6,2,3,0.01 g/L and pH 6.0, the highest xylanase activity reached to 21.15 U/mL, and increased by 79 % compared to the original enzyme producing medium.
