摘 要:本试验比较了乙醇提取、先乙醇后丙酮提取和先丙酮后乙醇提取3种蛋黄卵磷脂提取方法,以期获得一种最优的提取方法。同时,在各提取方法中分别采用不同乙醇体积分数观察其对蛋黄卵磷脂提取效果的影响。试验采用钼黄比色法测定卵磷脂的含量;通过薄层色谱法(TLC)定性检测蛋黄卵磷脂。结果表明:先利用95%的乙醇处理蛋黄,然后用丙酮脱油为最优蛋黄卵磷脂提取方法,卵磷脂的提取率、纯度和含量都较高,分别为8.6%,80.3%,6.9%。提取得到的蛋黄卵磷脂薄层色谱图和卵磷脂标准品的薄层色基本谱图一致。The aim of this study was to explore the best extraction method of egg yolk lecithin. The egg yolk was treated by ethanol, ethanol-acetone and acetone-ethanol organic solvent,and adopted different, ethanol volume fraction. The lecithin amount was measured by molybdenum- yellow-spectrophotometry method, and the property of lecithin was tested by thin-layer chro- matography(TLC) method. The results are as follows: The best procedure of egg yolk lecithin extraction was that dissolving the yolk with ethanol at first, and then deoiling with acetone. The extraction yield , lecithin purity and amount were 8.6 %,80.3 %, 6.9 %,respectively. The TLC chrom_atogram of extractive lecithin were as same as those of lecithin standard sample.
