摘 要:活性氧ROS是生理过程必要的代谢产物,适量的ROS起到信号转导作用,一旦过量会对细胞、蛋白质和DN A 造成损伤.在顺序注射-阀上实验室-化学发光系统内,优化并评价了鲁米诺-Co2+-ROS和鲁米诺-HRP-ROS 2种化学发光体系对ROS的检测条件和分析性能,并将其应用于肝细胞和植物叶片ROS的测定.结果表明,顺序注射-阀上实验室-ROS-化学发光检测法具有自动化程度高、分析速度快、重现性好和试剂试样用量少的显著特点,其中鲁米诺-Co2+-ROS 体系灵敏度高,相对发光强度与ROS浓度在0.5~1000 nmol · L -1范围内的回归方程为Y =49X+1700(R2=0.9924),检出限为0.27 nmol · L -1,对细胞和植物组织中 ROS 的回收率在95%~104%之间,结果令人满意.Reactive oxygen species (ROS) are physiological metabolites ,appropriate amount ROS play an essential role in signal transduction ,but excess ROS can damage cellular components such as pro-teins ,lipids ,and DNA .In this work ,employing sequential injection lab-on-valve chemiluminescent technique(SIA-LOV-CL) ,we explored and optimized the factors influencing Luminol-Co2+-ROS and Luminol-HRP-ROS chemiluminescent system ,respectively .We also applied the proposed method for the determination of ROS in Chang liver cells and plant tissue .The results showed that SIA-LOV-CL system for ROS determination not only possessed high sensitivity and stability ,but also minimized sample and reagent consumption .The relative CL intensity for Luminol-Co2+-ROS system was linear with ROS concentration in the range of 0 .5~1000 nmol · L -1 ,and a 0 .27 nmol · L -1 detection limit was achieved .The recoveries for the determination of ROS in Chang liver cells and plant tissue were in the range of 95% ~104% .
