摘  要:用于制浆造纸工业中的木聚糖酶要求纤维素酶活力很低 ,富含纤维素酶的木聚糖酶必须经过适当的纯化处理后才能使用 .提出了利用纤维性材料选择性吸附纤维素酶分离纯化木聚糖酶的方法 .研究表明 ,纤维素酶和木聚糖酶在纤维性材料上均存在吸附 ,但吸附性差异很大 .羧甲基纤维素 ( CMC)对纤维素酶具有强力的选择性吸附能力 ,在低温 ( 4℃ )条件下 ,当 CMC材料的用量为2 .5 g· L - 1时 ,通过间歇式吸附操作可以吸附去除原酶液 (木聚糖酶活力 2 8.3 U/ m L,CMC酶活力 0 .4U / m L )中 86.0 %的 CMC酶活力 ,而木聚糖酶活力的收率达 85 .5 % .通过 CMC柱子分离可以达到连续纯化木聚糖酶的目的 .The application of xylanases to pulp & paper industry requires them to be of quite low activity of cellulases; so cellulase rich xylanases must be treated with purification before being used. This paper presents a method of purifying xylanases—by the selective adsorption of cellulases on fiber materials. It was found that both cellulases and xylanases could be adsorbed on fiber materials, but they were quite different in adsorption degree. CMC had a strong ability to adsorb cellulases selectively. At low temperature (4℃),when CMC was used at 2.5 g·L -1 , 86.0% of CMCase activity in raw xylanases could selectively be eliminated, with 85.5% of xylanases activity recovered in the batch adsorption process. It was also found that xylanases could be purified continuously by CMC column.
