摘 要:目的确定重组Pichia pastoris GS115Muts菌株更适合的培养基和最佳接种时间。方法采用3种不同的培养基(YPD培养基、BMGY培养基和复合培养基)培养重组Pichia pastoris GS115Muts菌株,分别在4h和24h的时间间隔下,采用比浊法对酵母菌96h内的生长进行OD值测定。结果复合培养基和YPD培养基是3种培养基较为合适的,且在12~24h之间是重组酵母菌的对数生长期,此时是接种的最佳时间。结论合适的培养基和适当的接种时间对巴斯德毕赤酵母(Pichia pastoris)的培养关系密切。Objective This Experiment is testing the most suitable culture medium and the right time when the seed liquid is most applicable. Methods There are three kinds of culture medium in this experiment-YPD, BMGY, and compound culture medium. They are used to reform a new strain named Pichia pastoris GSl15Muts. The OD is tested every 4 and 24 hours, within 96 hours, in the way of turbidimetric method. Results The compound culture medium and the YPD are the better culture medium in three mediums. And the best time to vaccinate is the period between 12 and 24 hours. Conclusion Pichia Pastoris yeast strain is closely related to the suitable medium and appropriate inoculation time.
