酸性染料比色法测定玛咖总生物碱含量的方法研究

摘　　要：以苦参碱为对照品,采用酸性染料比色法测定玛咖总生物碱的含量,研究并建立玛咖总生物碱的测定方法。对检测方法使用的最大吸收波长、缓冲液pH、缓冲液用量、0.05%溴甲酚绿显色剂用量、溶液振摇时间、氯仿萃取时间进行了优化研究。结果显示,此方法精密度RSD=0.95%,回收率为108.03%,RSD=0.79%,表明该方法具有较好的精密度和稳定性;且操作简单,具有良好的可重复性。此方法测得云南丽江玛咖总生物碱含量为0.7%,苦参碱最低检测限为0.86μg。With the matrine as reference, establish a method for the determination of total alkaloids in Maca by acid dye colorimetry, and research and establish the determination methods of total alkaloids in Maca. The detection method of the maximum absorption wavelength, buffer pH, the dosage of buffer solution, 0.05% of bromoeresol green coloration agent dosage, solution of shaking time, chloroform extraction time were studied. The results showed that the RSD=0.95% of precision, the recovery was 108.03%, RSD=-0.79%, showed that the method had good precision and stability; And the operation is simple, with good repeatability. This method measured the content of alkaloids was 0.7% of Maca, the lowest detection limit ofmatrine was 0.86μg.
