摘  要:根据滚环扩增的原理设计引物、锁式探针以及反应体系,建立和优化了滚环扩增(Rolling Cycle Amplification,RCA)检测体系,构建可快速检测食品中的小肠结肠炎耶尔森氏菌的RCA检测方法,并进行了特异性和灵敏度试验。结果表明:所建立的RCA检测方法,具有很好的特异性,灵敏度为1.7×102CFU/mL,高于普通PCR方法。小肠结肠炎耶尔森氏菌RCA检测方法具有较高的灵敏度和特异性,适合在细菌快速检测方面应用。A new method, which based on rolling cycle amplification (RCA) to detect Yersinia enterocolitica of food sample, was established. The principle of rolling circle amplification technique was used to design primers, padlock probe and reaction system to establish rolling cycle amplification method. Specific and sensitivity were tested. The results showed that the sensitivity of RCA was 1:7 x 102 CFU/mL. Detecting Yersinia enterocolitica with RCA was more specific and sensitive than PCR method and has lower instrumental requirement. It's suitable for rapid detection of bacteria, with a broad prospect.
