摘  要:目的:观察过表达突变型SH3BP2(p.Asp419Gly)的单核细胞对颌骨成纤维细胞增殖的影响,探讨巨颌症病灶内纤维异常增殖的机制。方法:构建巨颌症致病基因SH3BP2突变(c.1256A>G)载体,转染单核细胞系Raw264.7,筛选过表达稳定株;以过表达野生型SH3BP2的Raw264.7稳定株为对照。制备Raw264.7条件培养基,培养颌骨成纤维细胞并对比其生长状态。结果:成功建立过表达细胞系。突变型组条件培养基中成纤维细胞减少速率小于野生型组(P<0.05)。结论:与野生型相比,突变型SH3BP2单核细胞对颌骨成纤维细胞生长抑制作用较弱,可能是巨颌症病灶常见纤维异常增殖的原因之一。Objective: To investigate the effects of monocytes over--expresson mutant SH3BP2 (p. Asp419 Gly) on the fibroblasts isolated from mouse mandible, for the purpose of exploring the possible reason of fibrosis in cherubism. Methods: We established the eukaryotic expression vector of mutant SH3BP2 (c. 1256A^G), which was transfected into the mus monocytes Raw264.7 to acquire the stable cell line over--expresson mutant SH3BP2 (p. Asp419 Gly); the Raw264.7 over--expresson wild type SH3BP2 were taken as control. Then the conditioned medium was collected to culture the fibroblasts isolated from mouse mandible, with which the growth was analyzed. Results: The stable cell lines of Raw264. 7 over--expresson wild type or mutant (p. Asp419 Gly) SH3BP2 were successfully established; and in contrast with the fibroblast cultured with the conditioned medium from wild type groups, the lost rate of fibroblasts with mutant groups was significantly lower (P〈0.05) . Conclusion: Compared to the wild type, the monocytes over--expresson mutant (p. Asp419 Gly) SH3BP2 exhibited less inhibition on the growth of mandible fibroblasts. Perhaps it results in the frequent fibrosis in cherubism at least partly.
