摘  要:目的评价临床实验室外周血淋巴细胞培养对染色体制备的影响,并建立高分辨染色体制备的方法。方法测量经秋水仙素分别处理1.75 h(常规方法)、1.5 h、1.25 h和1 h的外周血淋巴细胞的转化率及染色体分辨率情况,比较各实验组与常规方法是否存在差异。进行药物同步实验,计算细胞转化率,分析细胞染色体分辨率情况,比较药物同步方法与常规方法之间的差异。结果秋水仙素作用时间可减少至1.25 h,即可保证转化率及提高高分辨率染色体比例;药物同步方法可制备高分辨染色体,但存在转化率低、显带困难、实验效果不稳定等缺点。结论优化了外周血染色体分辨方案;建立了一套药物同步获得高分辨率染色体的方法,为提高小片段染色体异常病例的检出率提供基础。Objective To evaluate the impact of peripheral blood lymphocytes culture on chromosome preparation, and estab- lish high - resolution chromosome analysis. Methods The transformation rate and the chromosome resolution of peripheral blood lymphocytes were measured after colchicines treatment for 1.75 h (routine method), 1.5 h, 1.25 h and 1 h, then the difference between experiment groups and routine method was compared. In addition, the routine method was compared with the drug synchronous method by the transformation rate and the chromosome resolution. Results Colchicines treatment for 1.25 h can ensure the transformation rate and improve the proportion of high - resolution chromosome. The drug synchronization method can prepare high - resolution chromosomes, but it had the shortness of low transformation rate, difficulty in banding and instabil- ity. Conclusion This study was successful in getting more understanding of cytogenetic clinical work and optimization of the peripheral blood chromosome routine work; the preparation of high resolution chromosome by drug synchronization could be a good choice for examination of the small - fragment chromosome abnormality.
