摘 要:为了选育1株适于现代酒精工业发酵的高产酒精酵母菌新菌种,采用氯化三苯基四氮唑(TTC)筛选菌株并对其进行紫外线和亚硝酸复合诱变、最适发酵条件的单因素试验和正交试验,研究其发酵工艺。结果表明:选育得到1株稳定高产突变株UV-N-5,其产酒精度达8.25%(V/V),比初始菌株提高39.80%,并具有较好的耐高温和耐高浓度糖的特性;通过单因素和正交试验得到UV-N-5最佳的发酵条件为初始糖浓度15%、发酵温度35℃、起始pH 5、接种量8%、发酵时间72h;在该条件下,菌种UV-N-5发酵产酒精可达8.66%(V/V)。In order to breed a high-yield alcohol-producing yeast, TTC was used to screen bacterial strains, which were induced by UV and nitrous acid. And single factor test and orthogonal tests were conducted to optimize the fermentation conditions. Results: a stable high-yield mutant strain UV-N-5 was screened. Its alcoholic strength reached 8.25 % (V/V), which was 39.80 ~ higher than the initial bacterial strain and was of wonderful resistance to high temperature and high sugar concentration. The optimum fermentation conditions included initial sugar concentration 15~, fermentation temperature 35~C, initial pH value 5, inoculum size 8~ and fermentation time 72 hours. Under these conditions, the ethanol production could reach 8.66~ (V/V).
