摘 要:采用响应面法优化了从毛叶藜芦中提取环巴胺的条件。在单因素实验的基础上选取提取液料比,提取时间和提取温度为随机因子,以环巴胺提取量为响应值,建立三因素三水平Box-Behnken中心组合设计,并建立数学模型对响应值进行分析。结果表明,提取液料比、提取时间和提取温度对环巴胺的提取量都有极显著(p<0.01)的影响,并确定了最佳的提取工艺参数为:提取液料比9∶1(mL/g),提取时间10.0h,提取温度48.0℃。在此优化条件下,提取10.0g毛叶藜芦得到环巴胺的理论量为9.38mg,实际提取量为9.19mg,相对误差为2.02%。Response surface methodology was used to optimize the extraction conditions of cyclopamine from Veratrum grandiflorum. Based on single factor experiments ,the liquid-to-material ratio extraction time ,extraction temperature were chosen as causal factors and the extraction of cyclopamine was chosen as response value which was then analyzed by models. In this way ,the three-factors-three-Ievels Box-Behnken central composition experiments were designed. Results showed that liquid-to-material ratio, extraction time, extraction temperature had significant effects (p〈0.01 ) on extraction of cyclopamine. Optimal conditions for achieving the high extraction of cyclopamine were liquid-to-material ratio 9:1 (mUmg) ,extraction time 10.0h ,extraction temperature 48.0℃. Under these optimized conditions, with 1 0.0g raw materials extraction get cyclopamine 9.38mg in the abstract, the actual extraction of cyclopamine was 9.19mg,and the model prediction was worth 2.02 percent error.
