摘  要:目的研究放疗联合高效表达的胸苷激酶/丙氧鸟苷(TK/GCV)自杀基因系统对人肺腺癌细胞株A549细胞凋亡活性的影响,探寻肺腺癌放化疗联合治疗的新途径。方法利用前期研究成功构建的可调控自杀基因TK高效表达的pfos-iNOS/TK治疗载体转染A549细胞筛选阳性克隆。将阳性转染细胞(转染治疗载体组)和未转染的对照细胞(未转染组)分别给予2 Gy的X射线照射和10μmol/L、1000μmol/L的GCV。荧光显微镜观察细胞凋亡情况,检测各组细胞的凋亡率。结果给予10μmol/L、1000μmol/L的GCV作用后,治疗载体组凋亡率高于未转染(P<0.05);X射线照射联合GCV作用后,转染治疗载体组大量细胞发生凋亡,而未转染组仅有少量细胞发生凋亡(P<0.01)。结论放疗联合高效表达的TK/GCV自杀基因系统显著提高了对肺腺癌细胞的杀伤作用,为肺癌治疗带来了新的希望。Objective To explore a new way of radiochemotherapy for lung cancer by studying the effect of a suicide gene therapy system of thymidine kinase/ganciclovir(TK/GCV) and radiation on apoptosis of lung adenocarcinoma A549 cells. Methods A cancer cell line A549 was transfected by the vector pfos-iNOS/TK which was recombinated in previous study. The transfected cell and control cell were irradiated by X ray and 10μmol/L, 1000 μmol/L of GCV. Cell apoptosis was observed by fluorescence microscopy to detect cell apoptosis rate in each group. Results The ratio of apoptotic cell was higher in transfeeted group after therapy of 10 μmoL/L,1000 p.mol/L of GCV ( P 〈 0.05 ). With GCV and irradiation, higher apoptosis ratios were found in A549/pfos-iNOS/TK, while in control group only few apoptosis was observed ( P 〈 0.01 ). Condusion With radiation and more expression of TK/GCV, more A549 cells were killed and new hope is brought to the treatment of lung cancer.
