秸秆发酵产氢菌系的筛选及菌系功能强化研究  被引量:4

摘  要:采用连续稀释法从牛粪堆肥中筛选到一个可以较好的降解秸秆产氢的菌系CYY,该菌系的最适生长温度为37℃,最适pH为8.5,在秸秆培养基中,氢气产量为90 mL·L-1 culture。采用改良的Hungate厌氧滚管法,从菌系CYY中筛选到一株降解秸秆产氢效果较好的纯菌CYY-9,此菌为杆菌,最适生长温度为37℃,最适pH为8.5,利用秸秆产氢量为86 mL·L-1culture。将菌株CYY-9以一定比例添加到菌系CYY中,研究发现:当菌系培养到40 h左右时,以4%的比例接种菌株CYY-9,菌系的产氢能力最强,产氢量达到103 mL·L-1 culture,与原有菌系相比,产氢量增长了12.9%,秸秆降解率增长了49.2%,达到了对菌系生物强化的目的。Using the continual dilution method to screen a good strain CYY from the cow dung compost,which may produce hydrogen by degradation straw,the most suitable growth temperature was 37 ℃,the most suitable pH was 8.5.In the straw culture medium, the hydrogen output was 90 mL·L-1 culture. Using the improved Hungate anaerobic roll tube method to screen a pure bacteria CYY-9,that was bacillus,from strain CYY,which could produce hydrogen effectively by straw,the most suitable growth temperature was 37 ℃,the most suitable pH was 8.5,the hydrogen output was 86 mL·L-1 culture. strain CYY-9 was added to strain CYY by certain proportion,the research discovered that when the strain CYY raised about 40 h,the proportion with 4% inoculates with bacterial strain CYY-9,the new strain had the strongest hydrogen production ability,hydrogen amount reached 103 mL·L-1 culture. Compared with the original strain,the amount grew 12.9%,and the straw degradation rate grew 49.2%,had achieved the goal of strengthening the biological function of strains.
