摘　　要：根据植物乳杆菌ST-Ⅲ的基因序列设计多对菌株特异性引物,和细菌16S rRNA通用引物一起组合,筛选设计菌株特异性计数多重PCR。待测样品先用MRS琼脂平板进行培养计数,随机挑选一定数量的单菌落,提取DNA,做PCR鉴定。将传统平板计数技术与多重PCR技术有机地结合起来,初步建立了产品中植物乳杆菌ST-Ⅲ菌株特异性活菌计数方法。Multiple pairs of strain-specific primers for Lactobacillus plantarum ST- III were designed and synthesized according the complete gene sequence , and respectively combination with the universal primers of 16S rRNA to select the multiplex PCR for the strain-specific count. The bacterial DNA of the randomly selected colonies culture on MRS＇agar substrate were abstracted and identified by the multiplex PCR. This study tried to establish a method for counting the living bacteria of Lactobacillus plantarum ST- III in the product by using PCR and traditional plate count method technique.
