摘 要:为探讨黑曲霉B2所产β-葡萄糖苷酶对几种中药糖苷类化合物的转化能力,本研究通过硫酸铵盐析、DEAESephadex离子交换色谱等方法对黑曲霉B2所产β-葡萄糖苷酶进行了初步分离纯化,并以TLC法检测其酶解产物。结果表明,黑曲霉B2所产β-葡萄糖苷酶可在30 min内将各种中药糖苷类底物完全转化为相应苷元。表明该酶能够高效水解不同的中药糖苷类底物,具有较好的应用潜力。The conversion ability of β-glucosidase from Aspergillus niger B2 toward several glycosides in Chinese traditional medicine was investigated. The crude enzyme from Aspergillus niger B2 was subjected to (NH4) 2SO4 and DEAE-Sephadex anion-exchange chromatography, and the partially purified β-glucosidase was obtained. TLC detection of hydrolysis products by β-glucosidase showed that the enzyme could completely convert the glycosides to their aglycones within 30 min. It suggested that the β-glucosidase from Aspergillus niger B2 had a wide range of glycosidic substrates with high hydrolysis efficiency, hence having a good potential to be used in medicine production.
