摘 要:目的观察通过使用不同浓度黄芩素干预宫颈癌细胞后,Ras及CyclinD1 mRNA及蛋白水平表达变化。方法体外培养Hela细胞株,分为空白对照(control)组、100μmol/L干预组、200μmol/L干预组。培养24 h后,相差显微镜下观察细胞形态及数量的变化;流式细胞技术检测细胞周期的变化;RT-PCR法检测Ras及CyclinD1 mRNA水平的表达变化。Western Blot法检测Ras及CyclcinD1蛋白水平的表达变化。结果黄芩素干预24 h后,与空白对照(control)组相比较,各黄芩素干预组细胞周期G1期受到阻滞,阻滞程度随黄芩素浓度增加而增强(P<0.05);Ras及CyclinD1的mRNA及蛋白表达与黄芩素干预浓度浓度相关,呈逐渐降低的趋势(P<0.05)。结论黄芩素可通过抑制Ras信号通路,阻滞细胞周期G1期,影响细胞周期D1(CyclinD1)的表达,致宫颈癌Hela细胞凋亡。Objective To observe the changes of mRNA and protein expression of Ras and CyclinD1 in cervical cancer cells stimulated by baicalein. Methods HeLa cells cultured in vitro were exposed to baicalein (0, 100 and 200 μM) for 24 h. Then the periodic changes of each group cell was detected by flow cytometry;the mRNA changes of Ras and CyclinD1 were measured by PCR and protein expression of Ras and CyclinD1 were measured by western blot. Results Compared to control group, the cell cycle G1 of each group was inhabited (P〈0.05), baicalein significantly decreased the mRNA and protein expression of Ras and CyclinD1 in a concentration-dependent manner (P〈0.05). Conclusion Baicalein induces HeLa cell growth inhibition by inhabiting cell cycle G1, and by decreasing expression of Ras and CyclinD1.
