摘 要:【目的】研究外源肉碱对鲤鱼背肌脂肪酸组成及其CPT I基因表达的影响,为阐明肉碱调控脂肪酸代谢的机理提供理论依据。【方法】在基础饲料中分别添加0,50,100,150,200和400mg/kg L-肉碱,进行饲养试验,60d后取鲤鱼(Cyprinus carpio)背肌样品,测定其脂肪酸组成;提取鲤鱼背肌总RNA,采用同源克隆方法扩增CPT I基因的部分序列,Real-time PCR方法测定CPT I基因mRNA表达水平的变化。【结果】外源肉碱对鲤鱼背肌脂肪酸组成有显著影响(P<0.05),随饲料中L-肉碱添加量的增加,鲤鱼背肌∑SFA和∑MUFA脂肪酸含量显著升高(P<0.05),∑PUFA、EPA、DHA、∑n-3和∑n-6脂肪酸含量显著降低(P<0.05);同源克隆扩增获得了鲤鱼CPT I基因的部分cDNA序列(GenBank注册号为JQ361077),长度为857bp,同源性分析表明其与斑马鱼(Danio rerio)的同源性为89%。鲤鱼饲料中L-肉碱添加量为150mg/kg时,CPT I基因mRNA的表达丰度最高,为对照组的4倍。【结论】本试验条件下,外源肉碱添加量为150mg/kg时,显著影响鲤鱼幼鱼背肌脂肪酸组成和提高CPTI基因mRNA表达水平。[Objective] Effects of exogenous carnitine on fatty acid composition and CPT I gene expres- sion in muscle of common carp (Cyprinus carpio) were studied to better clarify the regulatory mechanism of carnitine on fatty acid metabolism. [Method] Six carp groups were fed with basal diet containing L- carnitine concentrations of 0,50,100,150,200, and 400 mg/kg for sixty days, and muscle samples were ex- tracted for detecting fatty acid composition and RNA were extracted for CPT I gene expression measure- ment using Real-time PCR. [Result] Fatty acid composition was significantly affected (P〈0.05) by L- carnitine. ∑SFA and ∑MUFA contents of carps fed with L-carnitine were significantly (P〈0.05) higher than those of the control group, while ∑PUFA, EPA, DHA, ∑n-3 and ∑n-6 contents were significantly (P〈0.05) lower. Partial cDNA of CPT I gene was cloned (GenBank accession number JQ361077) by ho- rnologous cloning, and the homologous alignment was analyzed. The obtained cDNA sequence was 857 bp in length,and its homology with zebra fish (Danio rerio) was 89%. 150 mg/kg L-carnitine resulted in the highest mRNA expression level, which was three times higher than that of the control group. [ConclusionI Adding 150 mg/kg L-carnitine to the basal diet significantly affected the fatty acid composition and CPT I gene expression level in muscle of common carp.
