摘  要:目的观察K-ras基因突变与乳腺癌术后复发的关系。方法提取149个乳腺肿瘤组织样本(设为观察组),选取同一患者正常乳腺组织样本(设为对照组),用聚合酶链式反应-单链构象多态性分析和直接测序法对K-ras基因突变情况进行分析,并探讨K-ras突变与乳腺癌术后复发的相关性。结果乳腺癌组织中K-ras突变发生率(45.64%)明显高于正常乳腺组织(2.01%)(P<0.05)。K-ras突变患者的术后3年累计复发率为79.41%(45/68),明显高于未突变患者的25.92%(21/81)(P<0.05)。结论 K-ras突变可能是导致乳腺癌术后复发的原因之一,可作为临床预后判断指标。Objective To investigate the relationship between K-ras gene mutation and the postoperative recurrence of breast cancer.Methods The 149 breast cancer tissue samples were extracted for the observation group, normal breast tissue samples of the same patients as control group. the K-ras gene mutation was analyzed by polymerase chain reaction single stranded conformation polymorphism (PCR-SSCP) and direct sequencing method. The correlation between K-ras mutation and the postoperative recurrence of breast cancer was investigated.Results Mutation rate of K-ras in breast cancer tissue (45.64%) was significantly higher than that in normal breast tissue (2.01%) (P〈0.05). Cumulative recurrence rate of K-ras mutation in patients with postoperative 3 year was 79.41% (45/68), significantly higher than that of patients without mutations(25.92%,21/81) (P〈0.05).Conclusion K-ras gene mutation is one of the reasons leading to a high postoperative recurrence in breast cancer patients. K-ras gene mutation can be used as a clinical prognostic indicator.
