摘  要:为弄清高黎贡山1株土壤淀粉酶的最佳产酶条件,采用摇瓶培养、发酵液作为粗酶液进行酶活测定,研究了不同培养基(酸碱)、不同温度和培养时间对淀粉酶产生菌产酶活性的影响。结果表明:碱性条件培养3 d、30℃下吸光值最大,即产酶最多;酸性条件培养4 d、50℃下吸光值最大,即产酶最多。该淀粉酶产生菌在碱性条件下可获最高酶活,达411.81 IU/(mL·min),而在酸性条件下可保持酶活稳定,为301.31~368.18 IU/(mL·min)。To explore the optimum conditions of amylase-producing strain in Gaoligong Mountain,the effects of different culture medium and different temperature and time on the production of amylase were studied by shake flask culture,with fermentation broth as a crude enzyme solution.Results:Under alkaline condition,incubation time of three days,30℃,the absorbance reached the highest,indicating the producing amount was the highest; and the absorbance reached the highest under acidic condition incubation time four days,50℃.The highest enzyme activity,411.81 IU/(mL·min),could be obtained under alkaline condition.Whereas the enzyme activity was stable under acidic condition,301.31~368.18 IU/(mL·min).
