摘 要:植物组织的扫描电镜样品干燥通常采用临界点干燥法和冷冻干燥法,但这些方法存在操作复杂,需要专门仪器等诸多不便。作者在工作中探索了六甲基二硅胺烷(HMDS)对样品进行干燥,并与冷冻干燥法做了比较。结果表明:在花期柱头和子房等幼嫩组织上,经HMDS干燥后的扫描结果较好且两种方法的结果几乎一致,在幼果期胚珠较为成熟的组织上,HMDS比冷冻干燥法的效果差。Samples of plant tissues for SEM are usually dried by methods of critical point drying and freeze-drying. However, there are limitations with these methods, such as complex operations, specialized equipment requirement and so on. In this paper, the samples were dried with Hexamethyl disilazane (HMDS). The effect was compared with those using the freeze-drying method. The results showed that samples in young tissues, such as the flowering stigma and ovary, could be dried well by both the HMDS method and the freeze-drying method and the effects of them on sample drying are similar. By contrast, the effects of HMDS on drying of mature ovule tissues were worse than that of the freeze-drying method.
