摘 要:利用响应面法对超声波辅助提取石榴籽多酚的工艺条件进行优化.在单因素试验的基础上选取乙醇体积分数、料液比、超声时间、超声温度4个因素为自变量,多酚提取量为响应值,采用响应面分析法,研究各自变量及其交互作用对多酚提取量的影响,建立二次多项回归方程预测模型.结果表明,超声波辅助提取石榴籽多酚的最佳工艺条件为:乙醇体积分数为34%、料液比为1∶24、超声时间为31min、超声温度为51℃、超声功率180W、超声频率80kHz.在该条件下,多酚类物质的实测提取量为7.73mg/g,与预测值仅相差1.69%.Response surface methodology (RSM) was applied in the optimization of process conditions for the extraction of phenolic compounds from pomegranate seed.On the basis of single-factor experiments,Box-Benhnken design involving four factors as independent variables was utilized to set up a quadratic polynomial regression equation for phenolic extraction yield as function of ethanol concentration,solid-liquid ratio,ultrasonic time,extraction temperature,and the respective variables and their influence on the yield were studied by using response surface analysis.The results showed that the optimal extraction conditions of polyphenol in pomegranate seeds assisted by the ultrasonic were:ethanol volume faction of 34%,the ratio of liquid-tomaterial 1 ∶ 24,ultrasonic time 31 min,extraction temperature of 51 ℃,ultrasonic power 180 W,ultrasonic frequency 80 kHz.The yield of polyphenols can be up to 7.73 mg/g under the optimal extraction condition,and the predictive value is only 1.69 % different.
