摘 要:为提高小麦麸皮内源性β–木糖苷酶提取得率,该文采用响应面法对其提取条件进行优化,在单因素实验基础上,选择提取液pH、料液比、提取时间为自变量,以β–木糖苷酶的酶提取量为响应值,根据Box–Benhnken Design法设计响应面试验,进行显著性和交互作用分析,确定酶的最佳提取条件并进行验证,结果表明:提取液pH 4.93、料液比1∶9.85 g/mL、提取时间36.85 min为最佳提取条件,酶提取量为84.19 mU/g,与响应面拟合优化预测值非常接近。In order to increase the yield of endogenousβ-xylosidase from wheat bran,this article optimized the extraction conditions ofβ-xylosidase by use of response surface methodology. Based on the single factor experiments,it designed the response surface experiments according to Box-Benhnken Design method by taking extraction pH,material-to-water ratio,extraction time as independent variables and the yield of endogenousβ-xylosidase as response value,carried out significance and interaction analysis to determine the optimum extraction conditions,and verified the results. The results showed that the yield of endogenousβ-xylosidase could reach 84.19 mU/g under the optimum extraction conditions of extraction pH 4.93,material-to-water ratio 1∶9.85 g/mL,extraction time 36.85 min,and the result was very close to the predicted value optimized by response surface fitting.
