摘 要:目的查明引起食物中毒的致病菌,分析菌株间的亲缘关系,为明确食物中毒诊断提供依据。方法采用定量PCR快速筛检、GB法分离、鉴定致病菌;用PFGE对病原菌进行同源性分析。结果从16份蛋糕标本中检出8株肠炎沙门菌,32份患者粪便标本中检出17株肠炎沙门菌,PCR核酸阳性与GB法分离到菌株完全一致;PFGE条带显示患者与蛋糕中检出的肠炎沙门菌带型一致,具有高度同源性。结论本起食物中毒由肠炎沙门菌污染蛋糕所致;PCR法与GB法联合检测有助于快速锁定食物中毒致病菌;PFGE检测发现蛋糕与患者检出的肠炎沙门菌其来源为同一克隆,从基因上证明了食物病原的相关性。Objective To find out the pathogens causing food poisoning,analyze the genetic relationships among strains and provide basis for clear diagnosis of food poisoning. Methods The pathogenic bacteria were screened with quantitative PCR, then separated and identified by GB method. The homology of the pathogens was analyzed by PFGE. Results Eight salmonella enteritidis strains were detected from 16 cake samples and 17 salmonella enteritidis strains were detected from 32 stool samples of patients,and the positive strains were consistent by PCR and the GB method. PFGE banding showed that salmonella enteritidis isolates from the patients had identical genotype with those from the cake,highly homologous. Conclusion This food poisoning was caused by salmonella enteritidis contamination in the cake. Combination of PCR method and the GB method was conducive to identify pathogen quickly in food poisoning. PFGE detection demonstrated the relevance of food and pathogens in terms of gene.
