摘 要:以微生物发酵床养猪技术带来的新兴农业副产物——微生物发酵床养猪垫料为主要培养基质,以产孢量为指标,通过单因素,响应面分析法优化生防菌哈茨木霉FJAT-9040固体发酵的培养基。单因素结果表明:发酵培养基主成份微生物发酵床养猪垫料与麸皮的比例为9∶1;最适疏松值、无机碳源和无机氮源分别为麦粒40.00%、蔗糖3.00%和硫酸铵2.00%。响应面分析结果表明,麦粒、蔗糖、硫酸铵对哈茨木霉产孢量存在显著相关性(p<0.05)。通过求解回归方程可得:当微生物发酵床养猪垫料∶麸皮=9∶1、麦粒含量36.74%、蔗糖含量3.07%、硫酸铵含量1.75%时,孢子产量最高,可达2.98×109个/g,与实际值相近。The optimization of solid-state fermentation culture medium for a biocontrol strain Trichoderma harzianum FJAT-9040 was studied using single factor analysis and response surface methodology (RSM). The medium mainly contained pig litter, which was produced from microbial fermentation bed for raising pigs, and the sporulation yield was used as the indicator in all tests. The results of single factor tests showed that the ratio of pig litter and wheat bran, the content of wheat grain, carbon and nitrogen were 9:1, 40.00% of wheat grain, 3.00% of sucrose, and 2.00% of ammonium sulfate, respectively. The results showed that wheat grain, sucrose and ammonium sulfate were significant for the spore of T. harzianum based on the response surface methodology analysis (2〈0.05). The optimal medium constitutes were as followed: 9:1 of pig litter and wheat bran, wheat grain 36.74%, sucrose 3.07%, ammonium sulfate 1.75%, and the average yield of spore reached to 2.98×109 cfu/g, which was close to the actual fermentation.
