摘 要:以实验室保存的印尼热泉水样和北极海水水样中的微生物为对象,通过平板划线、卢戈氏碘液染色和发酵上清液酶活测定等一系列定性定量相结合的方法,从两种极端环境水样中各筛选出4株具有高产卡拉胶酶能力的菌株,采用DNS法测上清液酶活来确定这8株菌所产卡拉胶酶的最适反应温度。结果发现,印尼热泉菌和北极海水菌所产的卡拉胶酶的最适反应温度分别为70℃、20℃。这两种酶分别对于高温和低温环境的工业生产具有非常大的应用价值。In this thesis,microorganisms in Indonesian hot spring water samples and in the Arctic water samples were studied as objects.Four strains with the ability of efficiently producing carrageenase were screened from the above two samples,respectively,in the combination of a series of qualitative and quantitative methods,including plate streaking,Lugol iodine staining and culture supernatant enzyme activity assay.After cultured at different temperatures,supernatant enzyme activity was measured by DNS method to study the optimum temperature of these carrageenases produced by the two kinds of extreme environment microorganisms.It was found that the optimum temperature of carrageenase from Indonesian hot springs bacteria and the Arctic seawater bacteria were 70℃and 20℃,respectively.These two kinds of enzyme have great application value in the industrial production under high or low temperature environment.
