摘　　要：目的探讨砷诱发永生化人表皮细胞(HaCaT)的恶性转化作用。方法 (1)长期低浓度亚砷酸钠(NaAsO2)处理HaCaT,观察细胞生长性状变化;(2)流式细胞术检测细胞周期;(3)注射处理后的HaCaT至裸鼠皮下致瘤试验。结果 (1)0.05 mg/L NaAsO2处理的HaCaT传代培养25代以后,细胞形态和生长方式均发生明显改变,增殖能力旺盛,倍增时间明显缩短;(2)0.05 mg/L NaAsO2处理的HaCaT细胞周期发生改变,处于S期的细胞比例增加;(3)0.05 mg/L NaAsO2所致恶性转化的HaCaT能在裸鼠皮下形成肿瘤;肿瘤组织病理切片显示为低分化鳞癌。结论长期暴露于低浓度NaAsO2可引起HaCaT发生恶性转化,且具有致瘤性。Objective To study the role of arsenic in inducing the malignant transformation of HaCaT cells . Methods ①HaCaT cells were treated with low concentration of NaAsO 2 for a long time and observed the changes of cell growth.②The cell cycle was detected by flow cytometry .③ The HaCaT cells were subcutaneously injected to nude mice ,and a tumorigenic experiment was carried out .Results ① The HaCaT cells showed obvious morphology and growth changes as well as strong proliferation and significantly shorter doubling time after the cells treated with 0.05 mg/L of NaAsO2 were subcultured to passage 25.② The cell cycle of HaCaT cells treated with 0.05 mg/L of NaAsO2 was changed,and the cells in phase S increased .③The malignant transformation of HaCaT cells induced by 0.05 mg/L of NaAsO2 caused tumor formation in nude mice .Tumor tissue pathology slices showed as low-differentiated squamous carcinoma .Conclusion The HaCaT cells might present malignant transformation being exposed to low concentration of NaAsO2 for a long time,which is of tumorigenicity .
