摘  要:目的:研究硒化乳酸菌胞外多糖对体外培养小鼠脾淋巴细胞分泌NO,IL-4及IL-12的影响。方法:分离制备了小鼠脾淋巴细胞,用Griess法检测NO含量,双抗体夹心酶联免疫法检测IL-4和IL-12含量。结果:当多糖质量浓度为400μg/mL时,硒化EPS对ConA刺激的T淋巴细胞NO分泌量高于EPS。培养4 h后上清液中NO浓度显著升高,培养8 h达到最高。硒化EPS可抑制小鼠脾淋巴细胞分泌IL-4,400μg/mL硒化EPS对于ConA刺激的小鼠T淋巴细胞IL-12生成量影响最为显著。结论:硒化EPS在一定程度上影响小鼠脾淋巴细胞免疫功能,这可能是其免疫调控机理之一。Objective: The effect of NO,IL-4 and IL-12 by selenium exopolysaccharides(EPS) produced by lactic acid bacteria were investigated in this study.Method: The concentration of NO in supernatant of vitro Spleen Lymphocyte was tested by griess assay,and the concentration of IL-4 and IL-12 was determined by Elisa.Results: It showed that 400 μg/mL Se-EPS group contains higher concentration of NO in supernatant of lymphocyte activated by ConA compared to other groups.After 4 hours,the concentration of NO in supernatant became much higher.The maximum NO production were observed after 8 hours.Selenium EPS can significantly inhibited the production of IL-4.A maximum IL-12 production were observed in lymphocyte activated by conA when the concentration of added selenium EPS in culture solution was 400 μg/mL.Conclusion: Se-EPS could effect the immune function of mouse spleen lymphocyte,which may be one of mechanisms.
