大杯蕈子实体多糖提取优化及抗氧化活性分析

摘  要:探析大杯蕈子实体多糖的适宜提取条件,明确其抗氧化活性.以大杯蕈子实体为材料,利用Plackett-Burman(PB)实验和响应面分析法(RSM)优化子实体多糖(FPS)的分离提取条件;以总还原力、羟基自由基、超氧阴离子自由基和二苯代苦味肼(DPPH)自由基为指标,测定FPS抗氧化活性.结果表明,FPS最佳提取条件是乙醇倍数3.55,醇沉时间28.77 h,提取次数2.57次,FPS理论得率是2.549%.为操作方便,调整为乙醇倍数3.6,醇沉时间29h,提取次数3.00,在此条件下,多糖的理论得率是2.329%,实际得率为2.015%.当FPS浓度为5000μg/mL时,大杯蕈子实体多糖的总还原力为0.629,对于羟基自由基、超氧阴离子自由基、DPPH的清除率分别为27.19%、60.101%和69.7%.随着FPS浓度的增加,其还原力逐渐增强,对羟基自由基、超氧阴离子自由基和DPPH的清除率也逐渐增大,证明大杯蕈子实体多糖具有潜在的抗氧化活性.[ Objective ] Clitocybe maxima belongs to Basidiomyeetes, Tricholomataeeae on the fungal sys- tematics. Clitocybe maxima has plenty of nutrition and significant function in medical health care. [ Method] The extraction parameters of fruting body polysaccharide (FPS) were optimized using Plackett - Burman test (PB) and response surface methodology (RSM) with clitocybe maxima. The FPS antioxidant activity were de- termined based on total reducing power, hydroxyl, superoxide anion and 1,1 - diphenyl -2 - picrylhydrazyl (DPPH) free radicals. [ Results ] The results showed that FPS optimal extraction parameters were as follows: ethanol multiple of 3.55, ethanol precipitation time of 28.77 h, extraction times of 2.57, and, The theoreti- cal value of FPS yield were 2. 549%. In view of convenient operation, the extraction parameters were adjusted to 3.6, 29 and 3. In this case, the theoretical yield of FPS were 2.329%, but the practical value was 2. 015%. FPS scavenging rate at the dosage of 5000 μg/mL on hydroxyl, superoxide anion and DPPH radicals were 27.19%, 60. 101%, and 69.7%, respectively, and its total reducing power was 0. 629. [ Conclusion] With FPS concentration increasing, its reducing power was enhanced and the free radical scavenging effects was amplified gradually, indicating that FPS had potential antioxidant activity.
