摘 要:目的:研究拟黄薄孔菌菌丝体的固体培养条件及胞内外过氧化氢酶(CAT)和超氧化物歧化酶(SOD)活性随培养时间的变化情况。方法:以菌丝生长速率及长势为指标探讨不同碳源、氮源、无机盐、温度、光照对菌丝生长的影响;采用紫外吸收法、氮蓝四唑(NBT)光还原法测定CAT、SOD活性。结果:拟黄薄孔菌菌丝生长的最适碳、氮源分别为葡萄糖、酵母粉,实验所选无机盐无明显促生长作用,最适温度25—35℃,连续黑暗和光暗交替均适宜菌丝生长;胞内CAT和SOD活力动态规律性不强,胞外不具CAT活性,胞外SOD总体呈先降后升的趋势。结论:初步建立拟黄薄孔菌的固体培养方法并观测其CAT和SOD随生长变化的情况。.[ Objective]To study the solid culture conditions of antrodia subxantha mycelium and test the CAT andSOD activity of the mycelium and fermentation liquor with it's growth period. [ Methods ] The effect of carbonsources,nitrogen sources,mineral salts,temperature and light on the mycelial growth of Antrodia subxantha wasresearched, according to the mycelial growth rate and the growth vigor.Ultraviolet absorption method and NBT lightreduction method were used to test the activity of CAT and SOD. [ Results ] The optimum carbon source wasglucose,the optimum nitrogen source was yeast powder, The mineral salts used in the trial do not promote themycelial growth obviously.The optimal temperature was from 25 to 35℃.It growth better in darkness entirely as wellas darkness with light alternately.The activity of CAT and SOD in mycelium had little regularity.CAT was not found inthe fermentation liquor.While the activity of SOD in the fermentation liquor increased after decreasing for a period.[Conclusions]Solid culture condition was founded initially.The activity of CAT and SOD with the processing ofculture was tested.
