摘  要:目的通过观察αA晶体蛋白基因启动子(αACP)与HSP70基因的融合基因表达载体(即pαACP-HSP70)对大鼠半乳糖性白内障的抑制作用,了解外源性HSP70在白内障发生发展中的作用。方法颈背部皮下注射D-半乳糖建立半乳糖性白内障大鼠模型;利用多价阳离子脂质体法包裹表达质粒,球后注射质粒到半乳糖性白内障大鼠眼部。裂隙灯观察大鼠晶状体混浊度的变化。通过荧光显微镜直接观察绿色荧光蛋白基因(EGFP)的表达。结果半乳糖+α-70组、半乳糖+HSP70组和半乳糖+EGFP组均可直接观察到大鼠晶状体上皮细胞中有绿色荧光,且半乳糖+α-70组荧光强度最强,与半乳糖+HSP70组、半乳糖+EGFP组有统计学意义(P<0.05),NS组与半乳糖+NS组未见绿色荧光蛋白表达;半乳糖+α-70组白内障出现的程度与同期的半乳糖+NS组、半乳糖+EGFP组及半乳糖+HSP70组相比明显减轻(P<0.05),并且其所出现白内障的时间亦延迟(P<0.05)。结论 pαACP-HSP70融合基因能在大鼠晶体上皮细胞内表达;外源HSP70可能起到延缓白内障发生和发展的作用。Objective To observe the inhibition effect of the expression vector(αACP-HSP70) on rat cataract induced by galactose and explore the role of exogenous HSP7 in rat cataract.Methods The model of cataract in rats was established by neck subcutaneous injection of galactose.The plasmids were parceled by polyvalent cationic liposomes and were injected into the rats by retrobulbar injection.Slit lamp was used to observe the lens turbidity.The expression of green fluorescent protein(EGFP) gene was observed by fluorescence microscope.Results ①The green fluorescence was obviously observed in the lens tissue of galactose + α-70 group,galactose + HSP70 group and galactose + EGFP group while not observed in the NS and galactose + NS group.The fluorescence intensity of galactose + α-70 group was strongest,which was a statistical difference compared with that of galactose + HSP70 group or galactose + EGFP group respectively(P 〈 0.05).②The degree of cataract in galactose + α-70 groups was reduced significantly comparing with the galactose + NS group,galactose + EGFP group or galactose + HSP70 group respectively(P 〈 0.05).Moreover,the time course of cataract development was proloned significantly(P 〈 0.05).Conclusion pαACP-HSP70 fusion gene can be expressed in rat lens and exogenous HSP70 can inhibit the development of rat cataract.
