活力素对荷艾氏腹水瘤(EAC)小鼠抑瘤作用的实验研究

摘　　要：目的探讨一种国家专利技术通过药食同源的"工程菌"转化含硒的中药灵芝孢子等提取多种中药活性成分(简称为活力素)的抑瘤及对抗肿瘤药的增效减毒作用。方法将活力素、环磷酰胺分别及共同用于荷艾氏腹水瘤小鼠,随机分为实验组(活力素组,环磷酰胺组,活力素+环磷酰胺组)及对照组,观察小鼠的存活天数,体重及瘤重变化情况。结果活力素组小鼠的存活天数明显延长,当活力素浓度为0.276g/mL时,能将小鼠生存时间延长24.4%;活力素组小鼠治疗后四周体重增加8.49g,明显高于对照组;环磷酰胺+活力素治疗组体重也有所增加,体重增加6.9g,略高于对照组;治疗四周后,将皮下瘤取出称重,发现活力素组肿瘤重量为0.531g,肿瘤小于对照组的0.686g,抑瘤率达22.6%,环磷酰胺组与环磷酰胺+活力素组肿瘤明显减小,抑瘤率更明显,分别为56.27%和76.68%。结论活力素能提高腹水瘤小鼠的生存时间,并对实体瘤的生长有明显的抑制作用,与环磷酰胺联合使用不但能明显提高抗肿瘤药物的抗肿瘤作用,还能减轻化疗药物降低体重的副作用,展示出与化疗药很好的协同抗癌作用。Objective To acquire some traditional Chinese medicine active components （named vigor） from transformed seleniumcontaining ganoderma spore by an engineering bacteria which is homology of medicine and food and investigate its anticancer effect alone and its efficacy enhancing and toxicity reducing effect in combination with other anticancer drugs. Methods Vigor and cyclophosphamide were applied in Ehrlich ascites carcinoma （EAC） tumor bearing mice alone and in combination,the mice were randomly divided into control group and experimental groups （inelucling vigor group, eyclophosphamide group and vigor ＋ eyclophosphamide group）, the survival days, body weight and tumor weight were observed. Results Survival days in vigor group were significantly increased,for example,when concentration of vigor was 0. 276g/mL,the survival days of mice were increased 24. 4%. After 4 weeks treatment, body weight in vigor group was markedly elevated 8. 49g compared with control group, whereas body weight in vigor ＋ cyclophosphamide group was slightly increased 6. 9g. Tumor weight in vigor group was 0. 531 g,less than 0. 686 g in control group, the tumor inhibition rate was 22. 6 %. Tumor weight in cydophosphamide group and vigor ＋ cyclopheaphamide group were significantly decreased with the tumor inhibition rate was 56. 27% and 76. 68% ,respectively. Conclusion Vigor could not only remarkably increase survival time of EAC tumor bearing mice,but also inhibit growth of tumor. In addition,combination of vigor and cydophosphamide could potentiate anticancer effect and attenuate side effect of chemotherapy drug,whieh showed anticancer synergy effect.
