大蒜素对胃癌细胞株及裸鼠移植瘤的抑制作用

摘　　要：目的　观察大蒜素对胃癌细胞株及移植瘤的作用 ,初步探讨抗癌机制。方法　采用MTT法观察不同质量浓度及作用不同时间大蒜素对胃癌 SGC- 790 1及 MGC- 80 3细胞的影响 ;流式细胞术分析 1 0 0 mg· L-1大蒜素分别作用 4h和 8h后两种肿瘤细胞增殖周期的变化 ;荷胃癌SGC- 790 1裸鼠模型随机分成 4组 ,每组 4只 ,分别于荷瘤 7d和 1 4d用 2 0 g/L的大蒜素 0 .1 m L和PBS0 .1 m L对瘤体进行局部注射治疗 ,每隔 2 d一次 ,共 3次 ,观察瘤体增殖及裸鼠生存状况的变化。结果　不同质量浓度梯度大蒜素对两种肿瘤细胞皆有一定的杀伤 ,大蒜素质量浓度为 80 0 mg· L-1及 1 0 0 0 mg·L-1时 ,对 SGC- 790 1细胞的抑制率分别为 65.5% ,68.6% (P<0 .0 1 ,vs5- FU)。大蒜素对 MGC- 80 3细胞的抑制作用较明显 ,质量浓度为 40 0 mg·L-1,80 0 mg· L-1及 1 0 0 0 mg·L-1时 ,抑制率分别为 67.5% ,75.3%和 79.5% (P<0 .0 1 ,vs5- FU)。 1 0 0 mg· L-1大蒜素连续作用3d,对 SGC- 790 1及 MGC- 80 3的抑制率分别可达 85.2 % ,96.7% (P<0 .0 1 ,vs5- FU)。大蒜素作用后 SGC- 790 1及 MGC- 80 3细胞 S期比例有增高趋势。荷瘤 7d局部注射治疗组有 3只裸鼠瘤体消失 ,余下 1只瘤体组织切片可见大量淋巴细胞聚集 ,肿瘤血管较少。Objective To study the anticarcinogen mechanism of garlicin after observation of the effect of garlicin on gastric cancer cells.Methods Cultural cell lines of gastric cancer SGC\|7901 and MGC\|803 were used as the target cells to observe the effect of garlicin of different concertritions with different culture time on these cell lines by MTT assay.Two kinds of gastrical cancer cells with 10% bovine serum and 100mg·L\+ -1 garlicin were cultured,and their changs of cell cycles after 4 and 8 hours were measured in vitro.In vivo study,garlicin were injected locally into the inplanted neoplasm of Balb/c nu/nu mice to observe the changs of the neoplasms and the living state after 7 and 14 days respectively when the nude mice tumor xenograft model were made.Results Garlicin had cytotoxic effect on these two kinds of gastrical cancer cells.When the mass concentritions of garlicin were 800mg·L\+ -1 and 1000mg·L\+ -1 ,the inhibiting rates of on SGC\|7901 cells were 65.5% and 68.6% respectively (P<0.01,vs5\| FU).When the mass concentritions of garlicin were 400mg·L\+ -1 ,800mg·L\+ -1 and 1000mg·L\+ -1 ,the inhibiting rates on MGC\|803 cells were 67.5%,75.3% and 79.5 respectively (P<0.01,vs5 \|FU).Culturing two kinds of gastrical cancer cells with bovine serum and 100mg·L\+ -1 garlicin for 3 days,the inhibiting rates could come up to 85.2% and 96.7% respectively.Culturing two kinds of gastrical cancer cells with bovine serum and 100mg·L\+ -1 garlicin for 4 and 8 hours,the S phase of the cell cycles rose,and that of the MGC\|803 cells rose more obviously.Injecting garlicin locally into the neoplasms in the seventh day when the nude mice tuor xenograft models were made,the neoplasm subsided in three nude mice,the neoplasm of one nude mice became very small.There were plenty of lymphocytes in the neoplasms,and the vasalium in neoplasm was scarce.Garlicin depressed the progress of the neoplasms when the nude mice tumor xenograft models were made by injecting locally in the fourteenth days,whose l
