摘  要:目的 观察共轭亚油酸 (conjugatedlinoleicacid ,CLA)对B16 MB细胞系侵袭能力的影响 ,探讨CLA预防肿瘤转移的可能作用机制。方法 用CLA 0、2 5、10 0、2 0 0 μmol L处理小鼠黑色素瘤B16 MB细胞系 ,观察CLA对人工合成基底膜的侵袭能力 ,对小鼠肿瘤细胞分泌明胶酶 (IV胶原酶 )的能力及肿瘤细胞表面钙粘蛋白 (E cadherin ,E CD)表达的影响。结果 CLA 5 0、10 0、2 0 0 μmol L明显降低B16 MB细胞系向基底膜的侵袭能力 ;CLA 10 0、2 0 0μmol L可抑制肿瘤细胞明胶酶的分泌 ,促进肿瘤细胞E CD的表达。 结论 CLA抑制肿瘤细胞的侵袭能力 ,其抗肿瘤转移作用可能与明胶酶分泌和EObjective To investigate the effects of conjugated linoleic acid on the invasive ability of tumor cells,and explore the possible pathway of the chemopeventive effects of conjugated linoleic acid on tumor metastasis.Methods B16\|MB cells were cultured in RPMI 1640 medium,10% fetal bovine serum and 0(control),25、50、100、200 μmol/L CLA for 24 hours and the ability of CLA\|treated cells to invade the reconstituted basement membrane was examined.At the same time,the level of type Ⅳ collagenases in RPMI 1640 medium and the expression of E\|CD in CLA\|treated cells were studied.Results At the doses of 50,100,200 μmol/L CLA,the invasion ability of B16\|MB through reconstituted basement membrane decreased.At the doses of 100,200 μmol/L CLA,the secretion of type Ⅳ collagenases declined and the expression of E\|CD increased.Conclusion Conjugated linoleic acid could inhibit the invasive ability of tumor cells and the mechanism of the anti\|tumors effects of conjugated linoleic acid may be associated with the changes of the secretion of type Ⅳ collagenases and the expression of E\|CD in tumor cells.
