两种不同的亲和吸附剂对猪凝血酶纯化效果的比较

摘　　要：以壳聚糖为载体、肝素为配基和以琼脂糖 4B为载体、肝素为配基分别合成 2种亲和吸附剂 ,用以纯化猪凝血酶 ,并对两者的纯化效果进行比较 .结果表明 :对于猪凝血酶的分离提纯 ,前者明显优于后者 .前者的酶比活为 1 5 5 3 .7U·mg- 1,提纯 1 1 .5倍 ,收率为 78.8% ;后者的酶比活为 1 1 2 6.4U·mg- 1,提纯 8.8倍 ,收率为 61 4 % .前者无明显压床 ,流速均匀 ,而且前者的价格低廉 ,易于得到 .Prepare two kinds of adsorbents, one was Chitosan\|Heparin adsorbent, the other was He\|parin\|Sepharose 4B adsorbent. Colums packed with two kinds of affinity adsorbent affinity chromatography were compared in the performance of porcine thrombin. The results showed the specific activity of thrombin purfied through Heparin\|Chitosan and Heparin\|Skpharose 4B was 1 553 U·mg -1 and 1 126 U·mg -1 , yield was 78 8% and 61 4%, purification fold is 11 5 and 8 8 respectively. The experimental results showed that Heparin\|Chitosan adsorbent is evidently better than Heparin\|Sepharose 4B adsorbent.
