香菇多糖对荷瘤小鼠IFNmRNA和SODmRNA的影响

摘　　要：目的 :通过研究香菇多糖 (L NT)对小鼠肝组织中干扰素 (IFN) m RNA和超氧化物岐化酶 (SOD) m RNA的影响 ,从分子水平探讨 L NT的药理作用机制。方法 :C5 7BL/ 6小鼠随机分成四组 ,其中三组左腋皮下接种 S1 80 肉瘤细胞。48h后 ,两组荷瘤小鼠分别腹腔注射 5 mg/ kg和 30 m g/ kg L NT,一天一次 ;另一组荷瘤小鼠及正常鼠等量注射生理盐水。 6天后杀鼠取肝 ,以斑点杂交的方法检测肝组织中 IFN m RNA和 SOD m RNA的表达量。结果 :5 mg/ kg L NT对 S1 80 肉瘤的生长有较明显的抑制作用 ;5 mg/ kg和 30 mg/ kg的 L NT均能提高 IFN m RNA和 SOD m RNA的表达量 ,且以 5 mg/ kg剂量组的作用稍强。结论 :L NT抗肿瘤和抗衰老的机理之一可能是促进了 IFN m RNA和 SOD mOBJECTIVE:Through the research on the impact of lentinan(LNT) to the IFNmRNA and SOD mRNA in mice liver,this artcle discussed the medicine mechanism of LNT on a molecular level.METHOD:We divided randomly the C 57 BL/6 mice into four groups.Three groups of mice were injected with S 180 cells (9×10 6 cells per mouse).After 48h,Two groups of S 180 bearing mice were injected with 5mg/kg and 30mg/kg lentinan respectively,and the other one S 180 bearing group and one model mice were injected with the same amount of 0.9%NaCl.Affer six days,kill the mice,weigh each S 180 and observe the content of IFNmRNA and SODmRNA in liver with dot blot.RESULTS:5mg/kg lentinan has a good inhibition on the growing of S 180 ;both 5mg/kg and 30 mg/kg lentinan can increase the content of IFN mRNA and SOD mRNA and 5 mg/kg lentinan has a stronger effect than 30mg/kg LNT.CONCLUSION:LNT could increase the content of IFN mRNA and SOD mRNA,which might be one of its mechanism against cancer and ageing.
