自杀基因疗法联合化疗对癌胚抗原表达型肺肿瘤的杀伤作用

摘　　要：目的　探讨阳离子脂质体介导E .colicd/HSV 1tk融合自杀基因联合长春瑞滨对癌胚抗原阳性肺癌的体内杀伤作用。方法　裸鼠皮下接种肺癌细胞SPCA 1后第 5、9、13天 ,腹腔注射阳离子脂质体 / pE CEAcd tk复合物 ,于第 6～ 15天连续腹腔注射 5 氟胞嘧碇 +丙氧鸟苷前体药物进行自杀基因治疗。于第 5、13天给予长春瑞滨进行联合治疗。结果　阳离子脂质体介导融合自杀基因联合长春瑞滨使小鼠皮下的肿瘤结节的生长受到显著抑制。其平均瘤体体积显著小于单独自杀基因或化疗治疗组的平均瘤体体积 (P <0 .0 1)。结论　阳离子脂质体介导E .colicd/HSV 1tk融合基因联合长春瑞滨化疗在体内可有效抑制肺肿瘤的生长 ,为癌胚抗原阳性肺癌的治疗提供了 1条新的途径。Objective To investigate the in vivo effect of Navelbine combined E.coli cd/HSV-1 tk fusion gene mediated by cationic liposome in anti-CEA positive lung cancer.Methods BALB/c mice were inoculated subcutaneously with SPCA-1 pulmonary adenocarcinoma cells and received intraperitoneal(i.p.)injection of cationic liposome DOSPER/ pE-CEAcd-tk complexes at the 5th,9th,13td days,followed by continuous injection with 5-FC and GCV prodrugs for 10 days from the 6th to 15th days.The tumor-bearing mice received combined treatment with suicide gene and Navelbine at the 5th and 13td days.Results A more significant reduction in tumor size was observed in the group of combination therapy compared with suicide gene therapy or chemotherapy alone.Conclusion Combined use of E.coli cd/HSV-1 tk fusion gene mediated by cationic liposome and Navelbine can significantly inhibit the growth of CEA posivitive lung cancer in vivo.It may be applicable for clinical gene therapy.
