摘 要:研究了用 13 98中性蛋白酶水解铬革屑提取水解蛋白质的各种影响因素 ,如酶的用量、反应温度和 p H值等对水解蛋白质收获率的影响。测定了水解蛋白质的灰分含量、铬含量等。综合各种影响参数 ,确定了制取水解蛋白质的最佳条件 ,在现有条件下制定了用碱和 13 98蛋白酶两步法处理铬革屑的最佳方案。研究结果表明 :采用此方案 Cr2 O3在水解蛋白质中的含量仅为 10 -6 级 ;水解蛋白质的回收率为 4 5 %左右 ,蛋白质总回收率达到 85During the past years,it was demonstrated that it is possible to digest the commercial chrome shavings-with an alkali in the first step and with an alkaline protease in the second step-to isolate two different protein products:gelatin and hydrolysate. In this study, the chrome sludge obtained from the first step was digested-with a neutral protease instead of the alkaline protease-to isolate the hydrolysate. Some parameters that affect the properties and yield of hydrolysate, such as the amount of the protease, pH,hydrolytic temperature and yield of hydrolysate so on, were studied. Also the properties of hydrolysate, such as yield, total ash and chromium content were measured. Finally, a set of optimum hydrolytic parameters was selected out. The results show that the process to treat the chrome shavings-with an alkali in the first step and with a neutral protease in the second step-to isolate gelatin and hydrolysate was a success.
