RGDS肽对TGFβ刺激肝星状细胞分泌细胞外基质的影响

摘　　要：探讨RGDS肽体外对TGFβ1刺激肝星状细胞分泌细胞外基质的影响。从正常大鼠肝脏中分离肝星状细胞原代培养 ,分 6组 ,单纯对照组 ,TGFβ1对照组 ,RGDS组 ,RGDS +TGFβ1组、RGES +TGFβ1组及IFNα - 2b+TGFβ1组。应用酶联免疫吸附测定方法检测各组培养上清中的Ⅲ型胶原。RGDS +TGFβ1组和IFN +TGFβ1组Ⅲ型胶原水平明显低于TGFβ1对照组、RGES对照组 (P <0 0 5 )。To probe into Arg-Gly-Asp-Ser(RGDS)peptide inhibiting hepatic stellate cells synthesizing extracellar matrix stimulated by transforming growth factor beta (TGF β). Hepatic Stellate cells (HSCs)were isolated from rat liver (male normal Sprague-Dawleyv rats; 550g)and plated onto 24-well uncoated plastic at a density 1×10 6 cells/ml and cultured. 6 groups were random divided which every group was 3 well cells. After cells cultured 72 hours, 100 μl cultured supernatants was respectively sucked from every well. Then, equality culturing medium was immited in the simple cultured cell control group, RGDS 100μg/ml was dropped into RGDS group wells, 100ng/ml TGF β 1 was only infused in simple TGF β 1 stimulating control group, and at one time TGF β 1 infused into other groups, 100μg/ml (final concentration) RGDS was infused in RGDS group and 100μ g/ml RGES in RGES group as well as 100U/ml interferon(IFN)α-2b in IFN group respectively, and they had been incubated together for 24 hours. Since, 100μl supernatants were taken from very well marked separately. The collagen Ⅲ level in the supernatants was detected by enzymelinked immunosorbent assay (ELISA). The collagen Ⅲ level of RGDS+TGF β 1 group and IFN+TGF β 1 group decreased significantly than of TGF β control group and RGES+TGF β 1 group (P<0.05). RGDS peptide can inhibit TGF β 1 accelerate hepatic stellate cells synthesizing collagen Ⅲ.
