摘 要:从大肠杆菌中大规模纯化新型干扰素 β(IFN βser17)。首先建立IFN βser17发酵和纯化工艺 ,连续大量纯化三批 ,并对纯化终产品进行全面鉴定。建立了稳定的发酵和纯化工艺流程 ,中试三批菌产量平均为 4.33g L ,IFN βser17的表达量平均为 2 0 % ,经破碎、粗提、精制后IFN βser17的比活达 2× 10 7IU mg以上 ,纯度超过 96 % 。The large scale fermentation and purification method were set up for IFN β Ser17, which was expressed in E. coli system. In the three batch of fermentation, the average harvest amount of cell was as high as 4.33 g/L of culture medium and more than 20% of somatic protein was expressed. The IFN βser17 was purified by centrifugation, ion exchange chromatography, etc. The purity reached 96% and the specific activity was more than 2.0×10 7 IU/mg. The other specifications also met the standard of the recombinant bio products of Chinese pharmacopoeia.
